What “99% Purity” Actually Means for a Research Peptide

I have a rule about numbers that everyone agrees on. When every vendor on the internet claims the same figure, the figure has stopped meaning anything. Peptide purity of 99% is the perfect example. It's on every product page, every banner, every Reddit vendor pitch. And almost nobody selling it can explain what it measures.

So let's do a myth-versus-fact pass on the most overused number in peptide marketing. Standard note first: these are research compounds, not FDA-approved drugs, sold for laboratory research only.

Myth: 99% purity means 99% of the vial is peptide

Fact: it doesn't. This is the single most common misunderstanding.

Purity, as measured by HPLC, is a ratio of peaks. The lab runs the sample through a column, the target peptide shows up as the main peak, and impurities show up as smaller peaks. Purity is the main peak's share of the total. It compares the peptide to the peptide-like junk that came along with it: truncated sequences, oxidized versions, leftover fragments from synthesis.

What HPLC purity does not measure is how much of the vial's total weight is peptide at all. Some of the mass is water. Some is counter-ions like trifluoroacetate. That's a separate number called net peptide content, and it's often noticeably lower than the purity figure. A vial can be 99% pure by HPLC and have a net peptide content in the 80s. Both can be true at the same time.

Myth: 99% pure means it's the right compound

Fact: purity and identity are different tests.

HPLC tells you how clean the sample is relative to related impurities. It does not confirm what the main peak is. That's the job of mass spectrometry, which measures the molecular weight of the sample and compares it to the expected weight for that peptide.

A 99% figure with no mass spec data next to it is a purity claim about an unidentified substance. I'd rather see 98.5% with a confirmed identity than 99.8% with no identity check at all.

Myth: the difference between 98% and 99% is trivial

Fact: it depends on what the other percent is, and you can't know that from the headline number.

Going from 98% to 99% cuts the impurity load in half. Whether that matters depends on what the impurities are. Some are harmless leftovers. Others are close cousins of the target peptide that can behave unpredictably in a research setting. The COA's HPLC chart shows how many secondary peaks there are and how big they are. That's more informative than the summary percentage, and it's why I always look at the chart, not just the number.

Myth: if the website says 99%, the batch is 99%

Fact: a website claim is not a test result.

Purity varies batch to batch. That's normal chemistry. A vendor who advertises "99%+" across the board is describing a target, not a measurement. The only number that counts is on the batch-specific COA for the lot you'd receive, with a lot number, a test date, and the name of the third-party lab that ran it.

If the vendor can't produce that, the 99% on the homepage is decoration.

Fact: what a real peptide purity 99 claim looks like

Here's what I want to see before I believe a purity figure.

  • An HPLC chart, not just a number, tied to a specific lot.
  • A mass spec result with observed and expected molecular weight.
  • A named third-party lab and a test date that makes sense.
  • Ideally, sterility or endotoxin results and net peptide content.
  • All of it available before I pay, not after I email support three times.

That last point filters out most of the market on its own.
